The Neural Stem Cell Biology Laboratory headed by François Guillemot

investigates how the fate decisions and behaviours of neural stem cells are regulated by extracellular signals and cell intrinsic transcriptional and epigenetic mechanisms.

We study neural stem cells in mouse embryos and in the brain of adult mice, as well as in human iPSC-derived neural cultures and foetal brains.

The laboratory uses research techniques of mouse genetics, 2D and 3D neural stem cell cultures, bulk and single cell investigations of the genome and epigenome of stem cells and their bioinformatic analysis (scRNA-Seq, scATAC-Seq, ChiP-Seq).

We are looking for a highly motivated researcher to study the transcriptional and epigenetic mechanisms that control the quiescent and active states in stem cells of the adult mouse hippocampus and the transitions between these states. This project builds on previous research from the Guillemot lab that has identified several stem cell populations in the adult hippocampus displaying different dynamics, and has discovered regulatory molecules that control the activity of these stem cells (Andersen et al. 2014, Neuron PMID: 25189209; Urban et al. 2016, Science, PMID: 27418510; Harris et al. 2021, Cell Stem Cell, PMID: 33581058).

Key Responsibilities

The post holder will work in the laboratory of Francois Guillemot and report to him as line manager. Postdoctoral Training Fellows are expected to lead their own projects, contribute to other projects on a collaborative basis (both in the lab and with external collaborators) and guide PhD students in their research. Two-ways scientific interactions will occur between the postholder and other members of the Guillemot group as well as with other laboratories including groups studying neural development and stem cells at the Crick. The ability to work in a team is essential. The successful applicant will use a wide range of techniques, including genetic, genomic and molecular tools.

In this project, some of the specific aims could include but not be limited to:

  • Characterising the chromatin accessibility profiles of different hippocampal stem cell populations using scRNA-Seq
  • Determining the roles of transcription factors ASCL1 and MYCN in establishing the transcriptional and epigenetic profiles of different hippocampal stem cell populations
  • Relating the age-related changes in hippocampal stem cell behaviour to transcriptional and epigenetic changes
  • Identifying transcription factors that act in combination with ASCL1 and MYCN to establish and maintain different hippocampal stem cell populations.  
  • To attend and report on research results at regular group meetings
  • To record all analyses in an accurate and clearly presented manner

Experience

Essential

  • PhD or in the final stages of PhD submission in biology or neuroscience
  • Significant experience in a laboratory (no prior postdoctoral experience is necessary)
  • Ability to work independently
  • Strong collaborative ability and teamwork experience
  • Strong communication skills and presenting ability
  • Strong organisational skills

Desirable

  • Experience with genomic approaches, e.g. scRNA-Seq/scATAC-Seq
  • Experience in bioinformatic analysis of large genomic datasets
  • Research experience in mouse genetics
  • Demonstration of scientific ability and excellence by means of high-quality refereed publications
