Toll-like receptor 7 (or TLR7) is an RNA sensor and immune-modulator that plays an important role in protection against viral infection. Brown et al. recently identified and described a single gain-of-function mutation in TRL7, Y264H, that causes the autoimmune disease lupus. The cover image shows a butterfly, the symbol for lupus (due to the typical “butterfly rash”), landing on the Y264H site of a flower grown from the protein structure of TLR7 (purple is the colour symbol for lupus).
