One of the fundamental questions in cancer evolution is the repeatability and stability of evolutionary trajectories.
Akin to the “rewinding the tape of life” though experiment proposed by Stephen Jay Gould using functional genomic approaches and patient-derived organoid models (organoids are 3D structure that consist of organ-specific cell-types that self-organize). We are testing how manipulation of the order of mutations - which is usually highly conserved - changes the cancer trajectory, potentially revealing therapeutic vulnerabilities.
